[COMPOSITION]
Each capsule contains: Rifampicin BP...300MG
[Appearance]
A practically odourless reddish brown or brownish red crystallin powder in capsules
[Pharmacology]
It is bactericidal against a wide range of micro-organisms and interferes with their synthesis of nucleic acids by inhibiting DNA- dependent RNA polymerase. It has the ability to kill itracellular or the spectrum of activity ganisms. The spectrum of actioity including Mycobacterium tuberculosis and M . leprea staphylococci,. Neisseria meningitidis, N. gonorrhoeae, Legionella spp. Rifampicin Chlamydia trachomatis and some anaerobic bacteria At high concentrations.it is active against some viruses. MICS for the most sensitive organisms(chlamydia, staphylo cocci)tend to range from about 0.01 to 0.02 ug per mL.while the MIC for most susc eptible mycobacteria ranges from 0.1 to 2ug per mL.
[Pharmacokinetics]
Rifampicin is readily absorbed from the gastro intestinal tract and peak plasma concentrations of about 7 to 10 ug pe mL have been reported 2 to 4 hours after a dose of 600 mg but there may be considerable interindividual variation. Approximately 80% wouldbound to plasma proteins. it is widely distributed in body tissues and fluids and diffuses into the cerebrospinal fluid when the meninges are inflamed an can crosses the placenta and distribute in breast milk. Half-lives for rifa mpicin have been reported to range initially form 2 to 5 hours. lt is ra pidly metabolised in the liver and excreted in the bile. Meanwhile up to 30% may be excreted in the urine ,
[Indication]
Rifampicin is used, notably is combination with isoniazid and pyrazinamide. as a component of multidrug regimens for the treatment of tuberculosis and with dapsone and clofazimine in the treatment of leprosy Other uses include treatment af staphylococcal infections, brucellosis, and legionnaires disease and the prophylaxis of haemophilus and meningococcal meningitis onnaues asease anatne propnylaxis
[Adverse Effects]
Rifampicin is usually well tolerated The main adverse effects area flu-like syndrome and a cutaneous syndrome. Hepatotoxicity also occurs G astro-intestinal adverse effects include nausea, vomiting, anorexia. diarrhoea, and e pigastric distress.
[Precautions]
Liver function should be checked before and during treatment, Blood counts should be monitored during prolonged treatment and in patients with hepatic disorders.
[Administration & dosage]
The usual adults dose of rifampicin is 600 mg daily by mouth. preferably on an empty stomach orally administration on an empty stomach in single daily doses of 10 mg per kg body-weight(maximum 600 mg) to children and adults is recommended. The maximum dose is considered to be 900 mg.
